Biotechnological methods are an essential component of plant genetic resources investigation that allow both to preserve a rare natural plant and useful selection genotypes, especially essential oil bearing plants used in medicine, perfume, food, etc. In the clonal micropropagation in vitro, the key moment is to retain genetic stability of in vitro material. It is considered that plant regeneration in vitro from meristem or vegetative buds gives identical clones, but the effect of the medium growth regulators on the genetic stability of in vitro plant material is discussed. Therefore, the objective of our work was to evaluate the genetic similarity of ex situ and in vitro plants. Investigation was performed on Hyssopus officinalis L. cv. Nikitskiy Beliy (NBG selection). Regeneration from the shoot single-node segments on the modified Murashige and Skoog culture medium (MS) supplemented with 6-benzylaminopurine (BAP) in concentration 0.3-0.9 mg/L and 0.1 mg/L Indole-3-butyric acid (IBA) was carried out. It was established, that application of 0.5 mg/L BAP was sufficient for the organ formation and development without morphological deviations. Genetic analysis based on RAPD and ISSR-PCR had shown full genetic similarity of the investigated ex situ and in vitro plants.

OBJECTIVES: Mumps is a highly contagious viral infection prevented by immunization with live attenuated vaccines. Mumps vaccines have proven to be safe and effective; however, rare cases of aseptic meningitis (AM) can occur after vaccination. The range of meningitis occurrence varies by different factors (strain, vaccine producer, and so on). Monovaccines or divaccines (mumps-measles vaccine), prepared from the strain Leningrad-3 (L-3), are used in Russia. Meningitis occurrence after vaccination has been established previously as very low. Nevertheless, with the number of children being vaccinated every year, vaccine-associated AM cases still occur. There is no official statistics on AM incidence after mumps vaccines, and information on AM features as an adverse event of mumps vaccination is limited and mostly devoted to vaccines, prepared from strains other than L-3. STUDY DESIGN: The study included patients with AM who were vaccinated against mumps in the previous 30 days before the present disease onset during 2009-2019. METHODS: Patients admitted to Infectious Clinical Hospital No. 1, Moscow, Russia, with AM were observed by a pediatrician and were screened for etiological agents of meningitis. RESULTS: Seven patients were enrolled, and clinical features and the course of infection are presented. CONCLUSIONS: Detection of only 7 cases of AM associated with mumps vaccination during the 10-year period supports very low occurrence of this adverse event after immunization with the L-3 strain-based mumps vaccines. Nevertheless, the annual number of AM cases that occur after mumps vaccination remains unknown and poorly diagnosed in practice because of the low awareness of physicians of this adverse reaction. Detection and objective coverage of such cases can lead to a weakening of 'antivaccination' moods in a society and to restoration of confidence in the healthcare system.

AIM: A pharmacoeconomic analysis of direct costs on treatment with high dose intravenous immunoglobulins (IVIG) and plasmapheresis (PP) in children. MATERIAL AND METHODS: Literature data on the pathogenesis of Guillain-Barre syndrome (GBS) were analyzed. The results of pharmacoeconomic analysis of direct costs on treatment of GBS using IVIG and PP are presented. Risks for complications during treatment with IVIG and PP are calculated. RESULTS AND CONCLUSION: The pharmacoeconomic analysis demonstrates comparable costs of treatment with IVIG or PP in the Russian Federation. Nevertheless, a less number of complications, convenience in use and the good safety and tolerability profile make it more preferable to this group of patients. In a clinical case of a 7-year child described in the article, treatment with 10% IVIG - privigen in dose 2 g/kg during 5 days started in the 3rd week of disease showed a marked positive effect.

We studied the effects of single administration of a suspension of magnetomicelles based on carbon-coated iron nanoparticles on the structure of rat lungs within 40 days. Histological analysis revealed a complex of hemodynamic alterations in the lungs. Described changes persisted in the lung stroma from day 1 until day 40, but their intensity decreased by the end of the experiment. Using immunohistochemical Perls reaction we identified cells morphologically corresponding to alveolar and interstitial lung macrophages. The number of Perls+ cells decreased by day 40 of the experiment. Ultrastructural analysis showed endocytosis of modified iron nanoparticles and their accumulation in intracellular digestionorganelles (endo- and lysosomes) of mononuclear phagocyte system cells. Accumulation of magnetomicelles in the lungs was not associated with damage to pneumocytes, macrophages, and blood-air barrier.

Two monoclonal antibodies (mABs) raised against plum pox virus (PPV) were shown to recognize its D, M, and C strains. Conjugates of the antibodies with colloidal gold (CG) nanoparticles averaging 26 nm in diameter were synthesized. The binding constants of PPV with both the native and conjugated mABs were determined using a Biacore X device. The complexes between the CG-mAB conjugates and plum pox virions were examined by means of transmission electron and atomic force microscopy. Using the conjugates with optimal component ratio, an express immunochromatographic assay of PPV was developed with a detection limit of 3 ng/ml and duration of 10 min. The assay was tested for PPV detection in samples of stone fruit tree leaves and demonstrated a good compatibility with the data obtained by "sandwich"-ELISA. The developed assay can be used in the field and applied for monitoring viral infection and for quarantine purposes.

RESUMO

Genome variability of in vitro micropropagated Clematis plants was established. The optimum concentrations of BAP and zeatin in the culture medium regulating in vitro morphogenetic processes in clematis explants cv. Serenada Kryma were determined. Molecular-genetic analysis of Clematis plants obtained via in vitro somatic embryogenesis and organogenesis was carried out. Using ISSR primers 105 amplicons have been revealed, six of them were polymorphic. The mean index of heterogeneity of clematis plants was 5.7%.
